The Church of Agia Sofia (Hagia Sophia, "Holy Wisdom") is a Byzantine church in the upper part of the medieval town of Monemvasia in the southern Peloponnese, Greece. It forms part of the town's wider archaeological site. The building was originally dedicated to Panagia Hodegetria, the Virgin Mary as guide. Over the centuries it served different communities under Venetian, Ottoman and Greek rule, which reflects the layered history of Monemvasia. It stands in the Laconia Regional Unit, near the square known as Plateia Elkomenou Christou.

The church was built in the upper town of Monemvasia and was first dedicated to Panagia Hodegetria. The Venetians, who held the town for a period, used it as a Catholic church dedicated to the Madonna.

During the Ottoman period the building was converted into a mosque. It was returned to Christian worship when Greece became independent in the early nineteenth century.
